Nanoindentation refers to a class of experimental techniques where micrometric force probe is used quantify the local mechanical properties soft biomaterials and cells. This approach has gained central role in fields mechanobiology, design tissue engineering, obtain proper characterization materials with resolution comparable size single cells (μm). The most popular strategy acquire such data employ an atomic microscope (AFM); while this instrument offers unprecedented (down pN) space (sub-nm), its usability often limited by complexity that prevents routine measurements integral indicators properties, as Young's Modulus (E). A new generation nanoindenters, those based on optical fiber sensing technology, recently popularity for ease integration allowing apply sub-nN forces µm spatial resolution, therefore being suitable hydrogels In protocol, step-by-step guide detailing procedure nanoindentation using commercially available ferrule-top nanoindenter presented. Whereas some steps are specific herein, proposed protocol can be taken other devices, granted adapted according manufacturer's guidelines. Further, open-source Python software equipped user-friendly graphical user interface analysis presented, which allows screening incorrectly acquired curves, filtering, computation contact point through different numerical procedures, conventional E, well more advanced particularly suited single-cell data.
